BRSK1wasinitiallyidentifiedasamammalianhomologtothefissionyeastS.pombeCdr2,amitosis-regulatorykinaseandalsoshowssignificanthomologytotheC.elegansneuronalcellpolarityregulatorSAD1.BRSK1isunbiquitouslyexpressed,withhighestlevelsofexpressioninthebrainandtestes.Similartoitsyeasthomolog,BRSK1isthoughttobeinvolvedinstress-inducedcellcyclearrest.OverexpressionofthisproteinleadstotheG2/MarrestinHeLaS2cellsandUV-inducedG2/MarrestcouldbepartiallyabrogatedbyreducedexpressionofBRSK1throughtheuseofsiRNA,indicatingitsroleinDNAdamagecheckpointfunction.Morerecently,ithasbeenshownthatbothBRSK1andtherelatedproteinBRSK2arerequiredformammalianneuronalpolarization.WhileBRSK1-andBRSK2-nullmicewereviable,double-mutantmicediedwithintwohoursofbirth.Neuronsfromthesemiceshoweduniformly-sizedneuritesasopposedtothenormallongaxonandmultipleshorterdendrites.TheseneuritesalsodisplayedbothaxonalanddendriticMarkers.AtleasttwoisoformsofBRSK1areknowntoexist.
